Genome sequences are foundation for functional and comparative,genomics studies.Two approaches with or without a physical map have been developed for whole genome sequencing.The map-based(clone-by-clone)approach is considered a gold standard but is laborious and costly,whereas the map-free(e.g.next-generation sequencing,NGS)approach has a high-throughput but has a huge problem in sequence assembly.
