Background Twin studies suggest that the establishment of substance use is strongly controlled by genetic factors.The short (s)/s genotype at the serotonin transporter gene linked polymorphic region had age-dependent effects on drug use.The short variant reduces the transcriptional efficiency of the gene resulting in decreased serotonin transporter expression in the neuron.Thus,expression of genetic vulnerability for substance use in children and adolescents may partly depend on interaction of serotonin transmission.Although serotonergic projections to dopaminergic pathways implicated in psychostimulant responses are believed to generally suppress their activity,studies examining serotonergic pharmacological manipulations on pyschostimulant actions have produced inconsistent results.In this study,we examined the net effect of raphe serotonergic neurons on cocaine responses using mice expressing"designer receptors exclusively activated by designer drugs (DREADD)".
