Oxidative stress plays an important role in the pathophysiology of cardiovascular diseases including hypertension, atherosclerosis, cardiac hypertrophy, and heart failure.Reactive oxygen species (ROS) are produced by multiple enzymatic sources located in the vascular wall.A family of NADPH oxidases appears to be especially important for redox signaling.In vascular smooth muscle cell, NADPH oxidases consist of Nox1, Nox4, p22phox, p47phox, and the small G protein Rac1.The NADPH oxidases are activated by agonists (including Ang Ⅱ, PDGF or IGF-Ⅰ) acutely (minutes), and are also chronically (hours) upregulated to further enhance ROS production.NADPH oxidases-derived ROS mediate many pathophysiological processes, such as cell growth, migration, and apoptosis.Much work remains to be done, however, especially in elucidating the precise mechanisms by which ROS modify the function of specific proteins.Our study demonstrated that insulin-like growth factor-Ⅰ (IGF-Ⅰ) enhanced ROS production in rat VSMCs.
