甘丙肽2型受体受泛素化调控的初步探讨

  甘丙肽(Galanin)是一个广泛分布于神经系统和外周组织中的神经肽。通过与其受体结合,Galanin参与调节认知、觉醒、摄食、生殖等生理过程。目前已克隆出甘丙肽受体的三种亚型,它们都属于G蛋白耦联受体(GPCR)。本研究在瞬时转染人甘丙肽2型受体(GalR2)的HEK293细胞系中给予蛋白酶体抑制剂MG132共同培养,发现GalR2在细胞中发生积累。这提示GalR2在降解过程中可能涉及泛素-蛋白酶体途径。在前人对GPCR的研究中显示,泛素修饰的GPCR既可能经蛋白酶体途径降解,也有可能作为分选信号调控囊泡的转运。在酵母双杂交的工作中,我们用人GalR2的C端作为bait,人脑cDNA文库作为prey筛选与GalR2-C端相互作用的蛋白。在酵母双质粒转化验证成功的相互作用蛋白中,有部分是与催化泛素化相关的蛋白。这些蛋白中除了典型的蛋白-蛋白相互作用区域之外,还都存在一个具有E3泛素连接酶特征的结构域,如SOCS-box,F-box等。这从另一个侧面也提示GalR2可能受泛素化调控。
