To constructed an allergen DNA vaccine Hum-CTLA-4-pcDNA3.1 comprising the main humulus allergen gene (EU088286) fused to CTLA-4 extracellular domain to improve the allergen to be administered directly to APCs.In this study Hum-CTLA-4 could induce Treg cells differentiation were certified.Further, we investigated that administration of the DNA vaccine protected the asthmatic mice from subsequent induction of allergic airway inflammation;Levels of IgE, Th2-type cytokines and eosinophil infiltration in Hum-CTLA-4-pcDNA3.1-administered mice were significantly lower than those in control plasmids-immunized mice, levels of Th1-and T-regulatory-type cytokines were enhanced by Hum-CTLA-4-pcDNA3.1 administration.The results of our study provide evidence for the potential utility of Hum-CTLA-4-pcDNA3.1 DNA inoculation as a novel approach for treatment of humulus-specific allergic asthma and other pollinosis.
