Sepsis,defined as the systemic host response to microorganisms in previously sterile tissues,is a syndrome related to severe infections and is characterized by end-organ dysfunction away from the primary site of infection.The normal host response to infection is complex,aiming to both identify and control pathogen invasion and start immediate tissue repair.Both the cellular and humoml immune systems are activated,giving rise to anti-inflammatory and proinflammatory responses.Morbidity and mortality remain unacceptably high despite increasing knowledge about the pathophysiological pathways and processes involved in sepsis and improved hospital care.Thymoquinone is the main constituent of the volatile oil of Nigella sativa.Animals (mice,n=12) weighing ~25 g were treated intraperitoneally for one-to-three days with thymoquinone prepared in 10% DMSO; a control group (treated with only 10% DMSO) was also simultaneously run.Four hours after last thymoquinone dose,groups were challenged with 2 mg/kg endotoxin (Lipopolysaccharide,LPS) or 2μl/g (1.5 × 107) Escherichia Coli (ATCC-25992) prepared in normal saline.Findings demonstrated a very potent protective activity against death (80-90% survival rate; P value <0.05) and improvement in biochemical and immunological parameters of sepsis including kidney and liver function tests,and tissue injury markers.In addition,significant reductions in the levels of inflammation cytokines including TNF-α,IL-1α,IL-2 and IL-10 (P values <0.05) were also observed.Thymoquinone could be a panacea for sepsis and a strong candidate for further development as a novel therapeutic agent for its management (Patent Pending).
