Purpose: To investigate the effect of penetration enhancers and ion pairing on the permeation of glibenclamide (GLB) through rat skin.Methods In vitro skin permeation experiment was performed using a side-by-side diffusion cell.Results All the penetration enhancers studied significantly increased the flux of GLB compared to the control and their enhancement activities were generally in the following order: menthol > N-methyl-2-pyrrolidone (NMP) > azone > oleic acid.The flux of GLB markedly increased in the presence of diethylamine (DEL), triethylamine (TEL),ethanolamine (EA), diethanolamine (DEA) and triethanolamine (TEA).The highest flux was observed with EA.Conclusion It is possible to enhance the flux of GLB by using an ion pair approach and the extent of enhancement possibly depends on the alkalinity, molecular weight, structure of the counter ions and the dielectric constant of the donor solution.
