高良姜素调节SIRT1表达诱导肝癌细胞自噬的研究

  目的:探索沉默信息调节因子1(silent information regulator 1,SIRT1)蛋白在黄酮类化合物高良姜素诱导肝癌细胞自噬过程中的调控作用,揭示SIRT1调控高良姜素诱导肝癌细胞自噬的分子机制。方法:培养HepG2细胞,分别设置对照组、抑制剂抑制SIRT1活性组、RNA干扰技术下调SIRT1表达组后,各组均加入35μg/ml高良姜素处理24小时,用MTT法检测各组细胞增殖情况,流式细胞术检测各组细胞凋亡情况,western Blot检测各组SIRT1、自噬相关蛋白和凋亡相关蛋白的表达情况,转染GFP-LC3荧光蛋白并用荧光显微镜观察各处理组的自噬泡情况,免疫共沉淀技术检测SIRT1和自噬相关蛋白家族ATG的结合情况以及ATG家族蛋白的乙酰化水平。
