Preparation and in vitro release of paliperidone oil suspension

　　OBJECTIVE:To sustain the release ofpaliperidone in vitro.METHODS Oil suspensions were prepared by mixing drugs with oil under vigorous vortex stirring.To investigate the effects of different factors,such as particle size,suspension medium,drug loading and dosage volume,on in vitro release.In vitro release studies of paliperidone oil suspension were performed in dialysis bags in wide neck flasks with 200ml of PBS (pH 7.4) at 37℃ in water bath oscillator.Dialyzate was withdrawn for analysis by HPLC at predetermined time intervals.RESULTS The optimal patch was acquired with drug loading of 6mg/ml,and dosage volume of lml.Soybean oil gave a slower drug release at early stage and higher release degree than sesame oil.Particle size had no significant effect on in vitro release within a certain range.CONCLUSION One simple preparation method for paliperidone oil suspension was established with steady in vitro release behavior for a week.
