目的:阐明大鼠巨噬细胞NR8383中TGF-β上调TLR2表达的分子机制.方法:用10 ng/mL TGF-β刺激NR8383细胞24小时后,用RT-qPCR和Western blotting检测TLR2 mRNA和蛋白质的表达;用Western blotting检测Smad4以及磷酸化的Smad2和Smad3的表达.结果:与对照组相比,10 ng/mL TGF-β刺激NR8383细胞后TLR2的表达显著增加,而且Smad4以及磷酸化的Smad2、Smad3也显著升高,提示TGF-β-Smad信号转导通路活化.结论:大鼠肺泡巨噬细胞NR8383中TGF-β通过转录因子Smad2、Smad3和Smad4上调TLR2的表达.
