Astragali Radix is widely used in daily foods as a tonic supplement.Hedysarum polybotrys is one species of Astragali Radix.In this study, the immunomodulatory properties ofH.polybotrys were explored by LPS-activated RAW 264.7 cells, splenocytes and daunoblastina-induced leucopenia in a BALB/c mice model.To monitor the quality of the H.polybotrys extracts, formononetin was used as a bioactive marker.RESULTS: H.polybotrys was extracted with hot-water and methanol, and then the MeOH extract was further partitioned with H2O and ethyl acetate (M-EA) to yield four fractions.M-EA had a richer formononetin content and stronger inhibited NO and PGE2 productions, iNOS and COX-2 expressions in LPS-stimulated RAW 264.7 cells and splenocytes than the other fractions.In addition, M-EA significantly stimulated proliferation of activated RAW 264.7 cells and splenocytes, scavenged NO radicals and prevented NO-induced cytotoxicity.Furthermore, M-EA also significantly increased the rate of the recovery of WBC level in daunoblastina-induced leucopenia mice.CONCLUSION: We examined M-EA stimulated RAW 264.7 cells and splenocytes proliferation through the action of scavenging NO radicals.H.polybotrys showed anti-inflammatory effects and regenerating immune cell popuiations of leucopenia mice.The scientific evidences proved the efficacy for health of this herbal supplement, H.polybotrys.
