天冬氨酸-β-CD衍生物拆分6种药物对映体

  本文合成了一种新的单一异构体离子型环糊精衍生物-天冬氨酸-β-CD衍生物(A-CD).以A-CD为手性选择剂,用毛细管电泳法对特布他林、氯丙那林、丙卡特罗、妥洛特罗、克伦特罗、卡维地洛6种手性药物进行了对映体分离研究.使用磷酸盐缓冲溶液为背景电解质,考察了手性选择剂浓度、背景电解质pH、缓冲盐浓度对分离的影响,优化了对映体分离条件.结果表明,在pH为2.0、浓度为50 mmol·L-1的磷酸盐缓冲盐中,添加12 mg·mL-1的A-CD,上述6种手性药物均能达到完全分离.研究表明天冬氨酸-β-CD衍生物对特布他林等6种手性药物具有很高的对映体选择性.
