目的:观察不同肿瘤细胞在放化疗抵抗中的作用及机制,探讨翻译调控肿瘤蛋白(Translationally controlled tumor protein,TCTP)和转化生长因子-β(transforming growth factor-beta,TGF-β)在此过程中发挥的作用.方法:实验选用人肠癌细胞SW480 和SW620、人肺癌细胞A549 和PC14、人乳腺癌细胞MCF-7 和MDA-MB-231,经2 Gy 60 钴(60Co)γ 射线照射.人乳腺癌细胞MCF-7 和MDA-MB-231,经化学药物5-氟尿嘧啶(5-FU)、盐酸阿霉素(ADM)及5-氟尿嘧啶+盐酸阿霉素(5-FU+ADM)诱导,药物浓度分别为5-FU:1-1.5μg/ml,ADM:0.08-0.15μg/ml;人胶质瘤细胞SHG44、U251 和U87 经化学药物替莫唑胺(TMZ)诱导,药物浓度为500μM-1000μM.
