The landscape phage demonstrating many excellent features including multivalency,stability,and high structural homogeneity,by which specific probes against targets can be screened from f8/8 displaying random octapeptides on the pⅧ coat protein of the phage fd-tet.In this talk,advanced phage clones with better specificity and affinity with targets for bioanalysis will be presented.First,specific probes for free prostate-specific antigen(fPSA)were screened and used as captured probe to establish a sandwich enzyme linked immunosorbent assay(ELISA)array of “phage/fPSA/anti-PSA mAb” combining with anti-PSA mAb,which is simple,cheap and efficient compared with immunoassay.In addition,novel peptide immobilization strategy based on symmetrical phage carrier possessing specific multivalent binding probes is proposed,which can dramatically increase the density and sterical regularity of immobilized peptides in each spot.Using this robust approach,we established peptide microarray for the detection of cellulase.The developed immobilization method based symmetrical carrier could circumvent the disadvantages of current peptide immobilization,such as the density,direction and composition.The suggested peptide immobilized method was efficient and reproducible,which was verified by comparing with the traditional method and quantitative model.Taken together,the landscape phage is an attractive,efficient and cost-effective biomolecular probe in bioanalyses.
