目的:建立大鼠实验性牙髓炎模型,了解Cryopyrin在大鼠牙髓组织的表达情况。方法:30只SpragueDawley大鼠随机分为0,1,3,5,7d5组。除0d组外,其余四组大鼠双侧下颌第一磨牙开髓后暴露于口腔环境中,分别于开髓后1,3,5和7d处死大鼠,分离双侧下颌骨。常规组织学处理后HE染色观察牙髓炎症状况,免疫组化检测Cryopyrin在牙髓炎中的表达及定位。结果:大鼠正常牙髓组织成牙本质细胞层Cryopyrin阳性表达,牙髓内部血管内皮细胞偶见表达,牙髓成纤维细胞未见表达。炎性牙髓组织血管内皮细胞表达增加,牙髓成纤维细胞及多种炎症细胞均有阳性表达,且表达的数量和牙髓的炎症浸润强度显著正相关(r=0.655,P<0.01)。结论:Cryopyrin在牙髓组织中表达并在牙髓炎的发生发展中起着重要作用。
Objective: To establish rat experimental pulpitis model to understand the expression of Cryopyrin in rat pulp tissue. Methods: Thirty Sprague Dawley rats were randomly divided into 0, 1, 3, 5 and 7 d 5 groups. Except for the 0d group, the other four groups of bilateral mandibular first molar were exposed to the oral environment after opening the pulp, rats were sacrificed at 1, 3, 5 and 7 days after the opening of the pulp, respectively, to separate the bilateral mandible. Histopathological examination was performed to observe the inflammatory state of pulp after routine histological treatment. The expression and localization of Cryopyrin in pulpitis were detected by immunohistochemistry. Results: The expression of Cryopyrin in rat dental pulp cells was positive, while that in dental pulp was occasionally expressed. There was no expression in dental pulp fibroblasts. The expression of vascular endothelial cells in inflammatory pulp tissue was increased, the number of dental pulp fibroblasts and many kinds of inflammatory cells were positive, and the quantity of expression was positively correlated with the inflammatory infiltration intensity of pulp (r = 0.655, P <0.01). Conclusion: Cryopyrin is expressed in pulp tissue and plays an important role in the pathogenesis of pulpitis.
