Polarized activities and roles of potassium channels in migrating hepatocellular carcinoma cells

　　K+ channels are involved in cell migration, but the mechanisms are unclear.It was demonstrated in this study by non-invasive micro-test and patch clamp techniques in migrating hepatocellular carcinoma MHCC97-H cells that: (1) knockdown of expression of the K+ channel IK1 by siRNA inhibited basal and volume-activated K+currents and cell migration;(2) K+ efflux was detected in the rear, but influx in the front;the K+ efflux, but not influx, was inhibited by the K+ channel blocker Clotrimazole;(3) H+ flowed out of cells in both poles, but the front efflux was stronger;(4) Ca2+ influx was detected only in the rear;(5) K+ channel activities were Ca2+-dependent and sensitive to extracellular pH.(6) IK1 was concentrated at the rear and ruffling edges, and was shipped dynamically from the front to the rear, detected by GFP-IK1 fusion technique.It is concluded that distribution and activities of the H+, Ca2+-sensitive K+ channel IK1 are polarized and modulated dynamically during migrating process, and play important roles in cell migration.
