The Molecular Pathology of Prostate Cancer in a Chinese-predominant Population

　　Prostate Cancer develops due to the interaction between cancer-related genes and proteins and the host microenvironment.The age-adjusted risk of prostate cancer (PC) in Singapore has increased since 1968.Genetic predispositions as well as an increasingly westernized lifestyle are likely risk factors for the disease incidence here.My presentation will cover the studies undertaken in our institute with respect to chromosomal abnormalities, genetic polymorphisms and over expression of androgen pathway genes and proteins in prostate cancer and their implications in the disease prognosis.We identified chromosomal copy number changes of important PC genes by fluorescence in-situ hybridization (FISH) showing significant association between gain ofc-myc and higher cancer Gleason scores and gain of chromosome 7 that was more frequent in locally advanced disease (pT3 stage).Genetic polymorphisms in the androgen pathway genes (androgen receptor, prostate specific antigen, 5-alpha reductase type Ⅱ and CYP 17) and their association with decreased PC and benign prostatic hyperplasia risk as well as over expression and association of androgen receptor, 5-alpha reductase isozymes, and vascular endothelial growth factor in PC will also be presented.
