Objective: To investigate the protective effects of different concentration of Ethyl Acetate (EA) extract from Chroogomphus on the degenerated dopaminergic neurons of PD mouse which induced by lipopolysaccharides (LPS).Methods: 30 Kunming (KM) Mice were randomly divided into five groups: the sham-operated control group, the LPS injection group, LPS+Ethyl Acetate Extract (100, 200, 400 mg/kg) group.Sterotaxic injection of LPS into mouse Substantia Nigra (SN) has been using to establish animal models of Parkinsons disease, and the immunohistochemical methods were used to observe the decrease of TH positive cells and morphological changes of glial fibrillary acidic protein (GFAP) in the substantia nigra (SN).Results: TH-positive neurons were ovale in the SN of LPS group, the neurite was shorter than control group, and the TH-positive cell numbers were significantly decreased in comparison with control group (P<0.05).Compared to LPS group, the TH-positive cells increased significantly in the LPS+100 mg/kg EA group (P<0.05) and LPS+200 mg/kg EA group (P<0.05).But there were no significantly difference among the LPS+100 mg/kg EA group, LPS+200 mg/kg EA group, and the control group.There was no difference of the TH-positive number between the LPS group and the LPS+400 mg/kg EA group.The GFAP expression was low-density in the control group, but in LPS group, the GFAP-positive cells density increased dramatically and the soma were bigger, the processes were longer.GFAP-positive cell numbers and density in LPS+100 mg/kg EA group and LPS+200 mg/kg EA group reduced Conclusion: The 100 mg/kg EA and 200 mg/kg EA group could protect the degeneration of dopaminergic neurons in SNc by depress the activity of astroglia cells in SNc, but 400 mg/kg EA group could not be considered to have the intervention effects.
