目的观察X线造影剂(RCM)对人血管平滑肌细胞的损伤及对其增殖的影响。方法(1)人血管平滑肌细胞种于24孔培养板上至长满培养孔,暴露于250mgI/ml泛影葡胺、低渗显影葡胺、优维显、伊索显及饱和甘露醇溶液15或60分钟,收集细胞并用台盼蓝染色鉴定细胞活性。(2)人血管平滑肌细胞暴露于250mgI/ml泛影葡胺、低渗显影葡胺、优维显、伊索显、饱和甘露醇溶液及10%胎牛血清(FCS)培养液15或60分钟后,用10%FCS培养液刺激细胞增殖,增殖开始前及增殖14天后作细胞计数。结果(1)暴露于250mgI/ml优维显、伊索显、饱和甘露醇溶液60分钟及250mgI/ml泛影葡胺、低渗显影葡胺15分钟后,细胞活性无改变,暴露于250mgI/ml泛影葡胺、低渗显影葡胺60分钟后分别出现16.5%±6.6%及9.2%±7.8%的死细胞。(2)250mgI/ml泛影葡胺、低渗显影葡胺及饱和甘露醇溶液抑制血管平滑肌细胞增殖,优维显及伊索显对血管平滑肌细胞增殖无影响。结论(1)高浓度离子型RCM有细胞毒性;(2)RCM对人血管平滑肌细胞的增殖无直接刺激作用;(3)RCM对细胞增殖的抑制作用与其高渗性和化学结构均有关
Objective To observe the effect of X-ray contrast agent (RCM) on human vascular smooth muscle cells and its proliferation. Methods (1) Human vascular smooth muscle cells were seeded on a 24-well culture plate to a full-length culture well and exposed to 250 mgI / ml diatrizoate, hypogonadal diatrizoate, prednisone, isohexidine, and saturated mannitol solution 15 Or 60 minutes, cells were harvested and cell viability was assessed by trypan blue staining. (2) Human vascular smooth muscle cells were exposed to 250 mgI / ml diatrizoate, hypogeminotriptyline, Uvoxil, Isoprost, saturated mannitol solution, and 10% fetal bovine serum (FCS) medium for 15 or 60 minutes After that, cell proliferation was stimulated with 10% FCS medium, before cell proliferation and after 14 days of cell proliferation. Results (1) The cells were exposed to 250mgI / ml of uvoxamine, isoprost, saturated mannitol for 60 minutes and 250mgI / ml of diatrizoate, hypoglycemic developing meglumine for 15 minutes without exposure to 250mgI / ml diatrizoate, hypogenetic diaberamine developed 16.5% ± 6.6% and 9.2% ± 7.8% of dead cells after 60 minutes, respectively. (2) 250mgI / ml diatrizoate, hypotonic hygroscopic solution and saturated mannitol solution inhibited the proliferation of vascular smooth muscle cells. Uvoxamine and Isoprost had no effect on the proliferation of vascular smooth muscle cells. CONCLUSION: (1) Cytotoxicity is observed in RCMs with high ionic strength. (2) RCM does not directly stimulate the proliferation of human vascular smooth muscle cells. (3) The inhibitory effect of RCM on cell proliferation is related to its hypertonicity and chemical structure
