目的 观察 75mGy、2GyX射线全身照射对小鼠脾淋巴细胞中IL 10的影响。方法采用Northernblot检测IL 10转录水平的变化 ,并同时通过流式细胞术检测其蛋白合成的变化。结果①蛋白合成结果表明 ,75mGyX射线全身照射后脾淋巴细胞IL 10合成在照后 2h开始即呈时间依赖性降低 ,至 48h仍无恢复迹象 (P <0 0 1) ,而 2Gy照射后则出现IL 10合成升高 ,于照射后 8h达到峰值 (P <0 0 5 )。②Northernblot结果表明 ,75mGy照射后脾细胞IL 10mRNA转录水平从照后 1h即降低 ,并维持较低水平一直到 48h开始恢复 ,达到假照射组的 88 35 %;2GyX射线全身照射后早期mRNA水平即有所升高 ,2h达到假照组的 134 0 6 %,4h略有下降 ,随后逐渐恢复至假照射组水平。结论 不同剂量X射线全身照射可引起不同的辐射效应 ,IL 10在其中起着重要的作用。
Objective To observe the effect of whole body irradiation with 75mGy and 2Gy X-rays on IL-10 in splenic lymphocytes of mice. Methods The transcription level of IL-10 was detected by Northern blot. Meanwhile, the protein synthesis was detected by flow cytometry. Results ① The results of protein synthesis showed that IL-10 synthesis of splenic lymphocytes decreased in a time-dependent manner from 2h after irradiation with 75mGy X-ray, but no signs of recovery were observed at 48h (P <0.01), but after 2Gy irradiation IL 10 synthesis increased, reached the peak 8 h after irradiation (P <0 0 5). ②Northernblot results showed that IL-10mRNA level in splenocytes decreased after 1h irradiation and maintained at a low level until 48h, reaching 88.35% of that in sham-irradiated group. The mRNA level of early stage after 2GyX-ray irradiation , Reached a high level at 134 0 6% in sham-operated group at 2 hours and decreased slightly at 4 hours, then recovered gradually to the level of sham irradiation group. Conclusion Different doses of X-ray irradiation can cause different radiation effects and IL 10 plays an important role.
